The advantage of this technique is that it can be used to detect exactly where a protein is located in the tissue under investigation, making it possible to detect specific contacts, cells or groups of cells. The procedures involve labelling the protein to be detected using specific antibody-antigen conjugates, which can then be detected using various microscopic techniques.

Binding at the cell surface or in the extracellular space can be labelled by a peroxidase-anti-peroxidase reaction, which forms a brown precipitate that makes the analyzed binding visible under light microscopy.

For fluorescence analysis of the tissue, a fluorochrome-linked antibody label is used, which is then detected by light of different wavelengths.

And to study the connections between neurons, we use immunogold labels of different sizes, mainly nanogold, which are then examined by electron microscopy.
