Generation of transgenic constructs and other type of expression vectors
Recombinant DNA laboratory offers diverse molecular biology services. Here, we plan and prepare the components necessary for the generation of genetically modified mouse lines. During the design phase, we analyze the gene or genes intended for modification or expression using bioinformatic methods to achieve optimal results. If the production of DNA constructs is required, we first assemble the necessary sequence elements in silico, order the components, and then proceed to create it step by step.
To expedite the work, gene synthesis plays an increasingly significant role in constructing sequences, although it is still not practical to produce the entire construct in this way.
We frequently perform the construction of Bacterial Artificial Chromosome (BAC) constructs, which is a useful tool when the goal is to replicate the expression pattern of an endogenous gene for transgene expression.link
In targeted gene modification, we primarily employ CRISPR/CAS technology. For deletions or smaller knock-in projects, the use of designed and synthetically produced components is usually sufficient. However, for larger knock-in projects, we also generate DNA constructs.
Our expertise extends beyond the creation of new mouse lines; we also produce constructs suitable for use in tissue cultures, such as expression vectors, as well as viral and non-viral constructs for somatic gene transfer and genome modification.
