Light and electron microscopic immunohistochemistry
Friday, 19 May, 2023
We use immunocytochemistry to determine neuronal or neuro-glial interactions and the localization of peptides/proteins in neuronal tissues.
Diaminobenzidine (DAB) and Ni-DAB chromogens are used for single-and double-labeling immunohistochemistry. Immunofluorescence combined with confocal microscopy is used for co-localization studies and for simultaneous localization of more than two peptides/proteins.
For ultrastructural localization of peptides/proteins, we combine electron-microscopy with single- or double-labeling preembedding immunocytochemistry using DAB, Ni-DAB chromogens and gold-colloids with or without silver intensification.
When necessary, tyramide signal amplification system is used to detect weak signals.
